dc.description.abstractBrazil has a high incidence of snakebite accidents, primarily involving snakes of the Bothrops genus, making necessary the search for more effective treatments with fewer side effects than the currently available antivenom serum. Literature studies show that in cases of envenomation, bothropic venom causes an intense inflammatory response and local and systemic hemorrhages, effects that may be related to proteolytic proteins of the venom, such as serine and metalloproteases (SVSPs – Snake Venom Serine Proteases and SVMPs – Snake Venom Metalloproteases). Although it is well established that bothropic envenomation causes clinical manifestations in sites distant from the bite, such as cardiovascular shock and acute renal failure, the implications of this condition on the male reproductive system need to be further investigated. Thus, this work analyzed the modulation of the testicular proteome of mice after experimental envenomation with Bothrops jararaca venom, as well as the effect of administering an endogenous metalloprotease inhibitor in this model. Using label-free LC-MS/MS, the results showed an increase in the abundance of proteins related to inflammation, apoptosis, and oxidative stress in the testicles of animals that received the bothropic venom injection. Proteomic analyses revealed that envenomation modulates the abundance of proteins associated with sperm motility and the regulation of male fertility, indicating possible impairment of testicular function. The obtained results indicate that the venom may disrupt the blood-testis barrier and dysregulate essential cellular processes, impacting tissue homeostasis and impairing male reproductive capacity. The administration of the endogenous metalloprotease inhibitor actinonin did not mitigate the effects caused by B. jararaca venom. Therefore, new studies are needed to characterize the action of this inhibitor in testicular tissue, for the development of therapeutic strategies that could be associated with bothropic antivenom for treating the systemic effects caused by snakebite accidents with B. jararaca. This is the first study in the scientific literature that verified, through the proteomic signature of testicular tissue in a murine model, the deleterious effects of bothropic envenomation on the integrity and functionality of male reproductive tissue.en
dc.description.resumoO Brasil apresenta alta incidência de acidentes ofídicos, principalmente por serpentes do gênero Bothrops, tornando necessária a busca por tratamentos mais eficazes e com menos efeitos colaterais do que aquele atualmente disponível, o soro antiofídico. Estudos na literatura mostram que, em casos de envenenamento, veneno botrópico causa intensa resposta inflamatória e hemorragias local e sistêmica, efeitos que podem estar relacionados a proteínas proteolíticas constituintes do veneno, tais como as serino e metaloproteases (SVSPs – Snake Venom Serineproteases e SVMPs – Snake Venom Metalloproteases). Embora seja bem estabelecido que o envenenamento botrópico cause manifestações clínicas em locais distantes da picada, como choque cardiovascular e insuficiência renal aguda, pouco se sabe sobre a implicação deste quadro no aparelho reprodutor masculino. Assim, este trabalho investigou a modulação do proteoma testicular de camundongos após envenenamento experimental com veneno de Bothrops jararaca, bem como o efeito da administração de um inibidor de metaloproteases endógenas neste modelo. Por meio de LC-MS/MS label-free, os resultados mostraram aumento na abundância de proteínas relacionadas à inflamação, apoptose e estresse oxidativo no testículo de animais que receberam a injeção do veneno botrópico. As análises proteômicas revelaram que o envenenamento modula a abundância de proteínas associadas à motilidade espermática e à regulação da fertilidade masculina, indicando possível comprometimento da função testicular. Os resultados obtidos indicam que o veneno pode romper a barreira hematotesticular e desregular processos celulares essenciais, impactando na homeostase tecidual e desregulando a capacidade reprodutiva masculina. A administração do inibidor de metaloprotease endógena actinonina não mitigou os efeitos causados pelo veneno de B. jararaca. Assim, são necessários novos estudos que caracterizem a ação desse inibidor no tecido testicular, para o desenvolvimento de estratégias terapêuticas que possam ser associadas ao soro antibotrópico para o tratamento dos efeitos sistêmicos causados por acidentes ofídicos com B. jararaca. Este é o primeiro estudo na literatura científica que verificou, por meio da assinatura proteômica do tecido testicular em modelo murino, os efeitos deletérios do envenenamento botrópico na integridade e funcionalidade do tecido reprodutivo masculino.pt_BR
